The effects of cytosine arabinoside upon proliferating epithelial cells.

چکیده
Experiments were conducted to obtain additional insight into the response patterns of the intestinal crypts to 1-0-D-arabinofuranosylcytosine, a potent inhibitor of DNA synthesis. The results indicate that this interference with nucleic acid metabolism is not lethally toxic to the crypt epithelial cells. This formulation is based on the following observations: (a) the absence of definitive ultrastructural manifestations of irreversible cell damage, particularly with respect to the nucleus; (b) the presence of labeled thymidine in virtually 100% of the mitotic figures occurring during recovery from the inhibitory effects of the analog; (c) the enhanced mitotic activity which closely followed the resumption of a considerable degree of DNA synthesis; (d) the failure to find any indication of intestinal epithelial cell death in animals killed 24 and 48 hr after the first of multiple injections of l-(3-D-arabinofuranosylcytosine; and (e) the absence of histolÃ3gica! evidence of cell death in a number of other epithelial tissues programmed for continuous replication. In contrast, however, stigmata of severe cytotoxic effects were readily discernible in the intestinal lymphocytes. It is therefore concluded that the apparent necrosis of the intestinal crypts noted with conventional microscopy actually represents phagocytosis of degenerating lymphocytic elements by adjacent crypt epithelial cells.

CYTOGENETIC EFFECTS OF MAGNETIC AND RADIOFREQUENCY FIEL DS OF NMRI SYSTEMS ON THE FREQUENCY OF CHROMOSOMAL ABERRATIONS INHUMAN LYMPHOCYTES IN THE PRESENCE OR ABSENCE OF CYTOSINE ARABINOSIDE
Application of nuclear magnetic resonance imaging (NMRI) as a non-invasive and accurate imaging procedure has been widely used in recent years. Meanwhile, the biological effects of magnetic fields of several tesla (T) and high energy radiofrequency (RF) is not fully known yet. Because of controversy over this issue, the present research has been carried out in order to verify the effects of...

The effects of methotrexate and cytosine arabinoside on polyamine metabolism were studied in a rapidly proliferating leukemic tumor (LI210) in C57BL/6 X DBA/2 (hereafter called BDP!) mice. Methotrexate, the most effective antileukemic drug in terms of increasing the median survival time also was the most effective, both in inhibiting the enzymes in the polyamine biosynthetic pathway and in decr...
متن کاملConcerning the necessary coupling of development to proliferation of mouse mammary epithelial cells.
The epithelial cells from mammary glands of mature virgin mice synthesize increasing levels of casein and cY-lactalbumin when cultured in the presence of insulin, hydrocortisone, and prolactin. When DNA synthesis is blocked in these cultures by the addition of cytosine arabinoside or 5-fluorodeoxyuridine, augmented milk-protein production does not occur. The effects of cytosine arabinoside are ...
متن کاملDepression of thymidylate synthetase activity in response to cytosine arabinoside.
Depression of thymidylate synthetase activity by cytosine arabinoside in CCRF-CEM cells was correlated with a blocking of precursor incorporation into RNA and with cell lysis. With increasing concentrations of cytosine arabinoside in the culture media, a proportional decrease of uridine incorporation into RNA and decrease in thymidylate synthetase activity occurred and was accompanied by lysis ...
